What our evidence ratings mean

What they did

Twelve adults whose depression had not responded to at least five previous treatments received a single 25mg dose of synthetic psilocybin, with therapy sessions before and after dosing.

What they found

Average depression scores fell by 17.2 points on the MADRS scale at twelve weeks. The five participants who also had PTSD improved considerably less than the rest.

Worth knowing

Open-label with twelve people and no control group, so this is a signal rather than a finding. The weaker response in the PTSD subgroup is the more useful detail here.

Every trial in this section used a single large dose, given in a clinic, with trained people in the room for the whole session. That is a different activity from anything sold on this site. More on why that distinction matters.
