What our evidence ratings mean

What they did

Adults with treatment-resistant depression across ten countries received a single dose of synthetic psilocybin at 25mg, 10mg or 1mg, each alongside psychological support, with depression measured on the MADRS scale.

What they found

The 25mg group improved 6.6 points more than the 1mg comparison group at three weeks. The 10mg group did not separate from the comparison.

Worth knowing

Side effects were common, with 77 percent of all participants reporting headache, nausea or dizziness. Suicidal ideation and self-injury occurred in the 25mg group. A single dose and a three week window is a short look at a chronic condition.

Every trial in this section used a single large dose, given in a clinic, with trained people in the room for the whole session. That is a different activity from anything sold on this site. More on why that distinction matters.
